◆反相层析
在吸附层析中，高极性物质在层析柱上吸附较牢，洗脱时发生拖尾现象和保留时间长的问题。如果在支持物上涂上一层高碳原子的疏水性强的烷烃类，洗脱液用极性强的溶剂，如甲醇和水的混合物。则被分离样品中的极性强的物质不被吸附，最先洗下来，得到较好的分离效果。这种层析法与普通的吸附层析法相反，故称为反相层析。目前用hplc做反相层析常用的ods柱，即在支持物的表面上连接了c18h37si—基团。
