固定化酶
immobilized enzyme

   用物理或化学方法处理水溶性的酶使之变成不溶于水或固定于固相载体的但仍具有酶活性的酶衍生物。在催化反应中,它以固相状态作用于底物,反应完成后,容易与水溶性反应物分离,可反复使用。固定化酶不但仍具有酶的高度专一性和高催化效率的特点,且比水溶性酶稳定,可较长期使用,具有较高的经济效益。将酶制成固定化酶,作为生物体内的酶的模拟,可有助于了解微环境对酶功能的影响。酶的固定化方法有载体结合法、交联法、包埋法等。酶经过固定化后,比较能耐受温度及pH的变化,最适pH往往稍有移位,对底物专一性没有任何改变,实际使用效率提高几十倍甚至几百倍。
   固定化酶的形式多样,可制成机械性能好的颗粒装成酶柱用于连续生产;或在反应器中进行批式搅拌反应;也可制成酶膜、酶管等应用于分析化学;又可制成微胶囊酶,作为治疗酶应用于临床。现在又有人用酶膜(包括细胞、组织、微生物制成的膜)与电、光、热等敏感的元件组成一种装置称生物传感器,用于测定有机化合物和发酵自动控制中信息的传递及环境保护中有害物质的检测。
