性质:是带有电喷雾离子化系统的质谱分析法。其原理如下:样品溶液经很细的进样管进入电喷雾室,在强电场的作用下,样品溶液在出口处因电荷的分离和静电引力而破碎成许多细小的带有电荷的液滴,在电场的作用下,带电液滴逆着干燥气体流动的方向,向质谱计入口处漂移,逆向的干燥气体使液滴迅速蒸发,并使液滴表面的电荷浓度增大,当库仑斥力和液滴表面张力极限值相等时,液滴就会爆裂成更小的液滴,直到液滴变得非常小,由于液滴的曲率半径很小,而它的表面电荷密度很大,结果在液滴表面形成非常强的电场,这电场足以从液滴中解吸出离子;离子经玻璃毛细管进入压力为几托(1托=133.322Pa)的第一真空区,在那里可以进行碰撞活化裂解,获得样品分子的碎片,从而可获得分子的结构信息。样品的分子离子和裂片离子经一系列分离器和静电透镜进入质量分析器进行质谱分析。这种电喷雾质谱法具有很高灵敏度,且电离的分子可以带有多电荷,这种多电荷离子的产生大大扩展了普通质谱仪能分析的质量范围,使质谱仪可以分析分子量为几十万质量单位的蛋白质分子。
