In this paper, used nickel metal ion chromatography and Sephacryl S-300 HR gel chromatography to purify Trehalose synthase from Thermobifida fusca the recombinant enzyme with a (His)_6tag at the amino terminus and investigate properties of it.
本实验利用镍金属亲和柱和Sephacryl S-300 HR凝胶柱层析纯化氮端带有6个组氨酸标签的来自褐色嗜热裂孢菌(Thermobifida fusca)的重组海藻糖合成酶并对其酶学性质进行了研究，实验结果表明：在肽链氮端引入六个组氨酸没有影响到该海藻糖合成酶的活性，1L的重组菌发酵液大概纯化出8。

在实验中，我们利用快速平板筛选法从鱼腹中获得了一株高产ChSase菌株YH311，并通过对菌株YH311进行生物学及生物化学等方面的鉴定，初步确定菌株YH311为温和气单孢菌(Aeromonas sobria)。

高温单孢菌属 细胞壁化学组份Ⅲ型，即以内消旋二氨基庚二酸为唯一特征性组份，无特征性糖。代表种为弯曲高温单孢菌，为嗜热菌，最适生长温度约50℃。
糖单孢菌属 形态上如高温单孢菌，但细胞壁化学组份Ⅳ型，即以内消旋二氨基庚二酸以及阿拉伯糖和半乳糖为特征性组分。代表种为绿色糖单孢菌。
