方法:以聚乙二醇6000为载体,采用溶剂法、熔融法、溶剂熔融法制备芦丁固体分散体,以芦丁溶出度(采用紫外-可见分光光度法)为指标成分考察制备时的冷冻时间;分别以聚乙二醇6000、聚乙烯吡咯烷酮、伯洛沙姆188(F68)、羟丙甲纤维素、β-环糊精、琥珀酸、乳糖等载体,仍采用三种常用方法制备固体分散体,以确定制备所选择的载体及方法;采用正交试验设计,以15min时芦丁溶出度为指标成分,优化制备的具体工艺;通过差示扫描量热法研究其物理化学性质的变化,为药物处方前的研究提供可靠的科学依据;比较固体分散体与普通物理混合物、芦丁原药加淀粉的溶出特性,并利用相似因子法进行统计学评价;研究不同温度、不同浓度的固体分散体的溶解度,从化学热力学角度探讨固体分散体的形成机制;最后进行固体分散体的稳定性实验,并初步建立胶囊的质量标准。
