性质:又称血管通透因子(vascular permeability factor)。1989首先从牛垂体滤泡状细胞培养液中分离纯化。有四种不同形式,成熟单体,分别为121,165,189和206个氨基酸,目前分离得到的VEGF,均由两个相同多肽链通过二硫键构成的同源二聚体,亚基分子量18 000~24 000,总蛋白分子量34 000~46 000。蛋白质表面有许多阳离子,呈强碱性,等电点高,对酸碱热稳定。VEGF是一类糖蛋白,可与肝素结合,多糖链仅与其分泌功能有关,与生物活性无关。VEGF 121/165为可溶性分泌蛋白。VEGF 189/206几乎都结合在质膜或基底膜上的含有肝素的蛋白聚糖上。四种VEGF的生物活性无明显差异。VEGF是一类多功能生长因子,能特异性作用于血管内皮细胞,强烈促进其增殖,促进血管形成,并有升高血管通透性的作用。多种细胞有分泌VEGF的功能,并通过旁分泌方式作用于血管内皮细胞。对血管内皮细胞的促有丝分裂作用十分强烈,可明显促进血管生成,能引起血管通透性增加,且是最强烈的引起血管通透性增加的物质。1nmol/L即可起作用,为组胺的50 000倍。用于心血管疾病、肿瘤、创伤修复等。
