产青霉素酶淋球菌(PPNG)检测
诊法。 因PPNG菌 株感染者日渐增多，青霉素治疗淋病疗效明显下降，故应检测PPNG。其方法为检测β-内酰 胺酶——纸片酸度定性法。步骤：滴上足量青霉素溶液(由结晶青霉素、溴甲酚紫及磷酸缓 冲液配制)。用接种环取10～20个淋球菌菌落，涂于滤纸上，约5mm直径，盖平四盖，置37℃ 温箱中30分钟，观察结果。产β-内酰胺酶的淋球菌株滤纸由蓝色变为黄色，通常于10分钟 内在滤纸上产生黄色斑点。机制：因其破坏青霉素中β-内酰胺酶环，形成青霉噻唑酸，使 试剂pH值降低，指示剂溴钾酚紫颜色由紫变黄。
