为了筛选开发新的活性物质,从舟山海域附近的海水、海泥、鲻鱼、斜带髭鲷的肠道中分离到120株菌株,以金黄色葡萄球菌、大肠杆菌、苏云金芽胞杆菌、黑曲霉、白假丝酵母为指示菌,采用双层琼脂法筛选到具有抗菌活性的菌株8株,其中菌株NF19表现出较好抗菌活性,对其进行16S rDNA序列分析,结果显示NF19是唾液乳杆菌。

性质：是最常用的纯种分离方法。先将待分离的材料作一系列的稀释（如1：10，1：100，1：1000，1：10000……），然后分别取不同稀释液少许，与已融化并冷却至45℃的琼脂培养基相混合，摇匀后倾入灭过菌培养皿中，待琼脂凝固后，保温培养一定时间即可出现菌落。如果稀释得当，平皿上就可出现分散的单个菌落，这个菌落可能是由一个细菌繁殖形成的。随后挑取该单个菌落，重复上面操作数次，便可得到纯培养。
