In 1893, Biginelli reported the first synthesis of 3,4-dihydropyrimidin-2(1H)-ones (DHPM) by a very simple one-pot condensation reaction of an aromatic aldehyde, urea and ethyl acetoacetate.

性质:无色结晶。熔点235℃(分解)。溶于热水、醇、醚、碱及碳酸钠溶液,微溶于水。

制备方法:由丙二酸二乙酯与硫脲反应而得。将硫脲溶于甲醇中,加入甲醇钠及丙二酸二乙酯,加热回流4-5h,然后减压蒸馏回收甲醇至尽,冷却后加入盐酸酸化至pH为1-2。放置过夜,析出硫代巴比妥酸结晶,过滤,用水重结晶即为纯品。

用途:用作电镀添加剂、生化试剂、铑的分析试剂和药物中间体。
