In order to establish a stable ISSR-PCR reaction system of Scrophularian ningpoensis Hemsl,some compatible PCR systems were screened by orthogonal design on 5 factors(dNTPs,Taq DNA polymerase,MgCl2,primer and DNA template) and 5 levels,meanwhile the factors and reaction procedure(annealing temperature,elongate time and cyclic numbers) were optimized by single factor gradient.
为了建立稳定的玄参ISSR-PCR反应体系,本试验采用5因素(dNTPs、Taq DNA聚合酶、MgCl2、引物及模板)5水平正交设计筛选PCR体系,并利用单因子梯度进一步优化反应体系各因素浓度和扩增程序(退火温度、延伸时间和循环次数)。

Base on the invariable quantity of TaqDNA, we att empted to establish and optimize ITS-PCR amplification system for Buxus sinica var.
本研究利用改良CTAB法从珍珠黄杨叶片中提取基因组DNA作为模板,在TaqDNA聚合酶量不变的基础上,利用正交设计L9(34)对4个因素(模板DNA、Mg2+、dNTP和引物)在3个水平上对珍珠黄杨ITS-PCR反应体系进行优化。
