性质:药物代谢动力学参数随剂量(或体内药物浓度)而变化的代谢过程如生物半衰期与剂量有关,又称剂量依赖性动力学。它具有以下特点:药物代谢不遵守简单的一级动力学过程,而遵从Michalis-Menten方程;药物的消除半衰期随剂量的增加而延长;血药浓度-时间曲线下面积(AUC)与剂量不成正比,当剂量增加时,AUC显著增加;平均稳态血药浓度也不与剂量成正比。
