There are two parts in this study:the first part is the clone ofpheophorbide a oxygenase(PaO) gene from wheat spike of photoperiod-and/or temperature sensitive male sterility wheat line BS210 based on RT-PCR and TAIL-PCR techniques and the expression profiles of PaO gene was analyzed in spike tissues of BS210 line cultivated in different fertile conditions.
本研究克隆2个基因,分别是小麦脱镁叶绿(甲酯)酸加氧酶(pheophorbide a oxygenase,PaO)基因和Ca~(2+) -ATPase基因,应用RT-PCR结合TAIL-PCR技术克隆PaO基因的cDNA全长,并利用半定量RT-PCR研究了该基因在小麦光温敏雄性不育系BS210穗部不同育性生长期的表达特性,并构建了植物表达载体(双子叶植物和单子叶植物)。

The three complete LIPOR genes,chlL,chlN and chlB,were PCR-cloned from the green alga Chlorella protothecoides CS-41(CSIRO),and these chloroplast genes showed very high identity(>99%) with the corresponding genes in Chlorella vulgaris C-27 chloroplast according to the results of sequencing and sequence alignment.
