For the passed decades, the synthesis of 4-aminopyridine has been performed by the preparation of pyridine-N-oxide from the oxidation of pyridine using H2O2 as oxidant in the presence of acetic acid, and then 4-nitropyridine-N-oxide using the concentrated sulfuric or nitric acid, followed by the reduction using iron powder in the solvent of acetic acid.
目前国际上常用的合成方法是在大量乙酸存在下用30%的H2O2氧化吡啶制备吡啶-N-氧化物,以硝硫混酸为硝化剂硝化得到4-硝基吡啶-N-氧化物,再以铁粉为还原剂在乙酸中还原得到4-氨基吡啶。
