本研究以新鲜甲鱼作为蛋白质底物,采用二次回归正交旋转组合设计对胰蛋白酶、木瓜蛋白酶和中性蛋白酶水解甲鱼蛋白的参数进行优化,建立了甲鱼蛋白水解度(Y_a)、蛋白质回收率(Y_b)与酶用量(X_1)、酶解温度(X_2)、pH值(X_3)及酶解时间(X_4)四个试验因素的正交回归模型;以蛋白质水解度和蛋白质回收率为指标,比较了胰蛋白酶、木瓜蛋白酶及中性蛋白酶水解甲鱼蛋白的效果,并分析了甲鱼蛋白酶解前后游离氨基酸的变化;在此基础上,进行了乳酸菌对甲鱼蛋白木瓜蛋白酶水解液发酵技术参数的优化。
